Protective Effects of Eggplant (Solanum melongena L.) against Oxidative Stress under In Vitro and Cellular System

Abstract
The protective effects of eggplant (Solanum melongena L.) against oxidative stress were investigated under in vitro and cellular system using LLC-PK1 renal epithelial cells. The eggplant were extracted with methanol (MeOH) and then fractionated into three different layers: chloroform (CH2Cl2), butanol (BuOH) and aqueous layers(H2O). The extract and fractions of eggplant showed strong 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity. In particular, among the fractions, BuOH fraction exerted the strongest effect with the IC50 value of 4.53¥ìg/ml. In addition, the fractions except for CH2Cl2 also showed nitric oxide (NO) scavenging effect. The BuOH fractions showed superoxide anion (O2
?) radical scavenging activity. Even though it was not concentration- dependent manner, BuOH fractions at the concentration of 1 mg/ml showed O2 ? scavenging effect of 44.01%. It also exerted strong hydroxyl radical scavenging activity. Moreover, under LLC-PK1 cellular model, the BuOH fraction ameliorated oxidative stress induced by 2,2¡¯-azobis (2-amidinopropane) dihydrochloride, the generator of peroxyl radical. In addition, the protective effect of BuOH fraction from NO under cellular oxidative stress was also studied. The LLC-PK1 cells showed the decreases in cell viability by the treatment with generators of peroxynitrite, 3-morpholinosydnonimine, and its precursors NO and superoxide anion produced by sodium nitroprusside and pyrogallol, respectively. However, the BuOH fraction led to the significant
increases in the cell viability with dose-dependence. The present study suggests that eggplant would have the protective activity from radical-induced oxidative stress and its BuOH fraction would play the crucial role.
